That's rude.

You can't request that the seat next to you remain empty.

You can hope for that, and you can switch your seat at the last minute to a seat with an empty seat beside it, if that's available.

But there are no guarantees.

I've read that even if somebody actually books and pays for the seat next to them, flight attendants will sometimes sit another passenger there if the flight is overbooked.
